What is Cancer Stem Cell Biology?

Cancer stem cells (CSCs), also called tumor-initiating cells, are a subpopulation of tumor cells with the capacity for self-renewal, differentiation, and tumor reconstitution. They are implicated in therapy resistance, dormancy, recurrence, and metastasis.

CSCs are identified by surface markers (such as CD44, CD133, ALDH activity), functional assays (sphere formation, serial transplantation), and gene expression signatures. They are maintained by signaling pathways including Wnt, Notch, Hedgehog, and Hippo, and they often inhabit specialized niches.

Therapeutic strategies targeting CSCs include inhibitors of stemness pathways, differentiation therapies, immune-mediated approaches, and combinations with conventional treatments. Identifying robust CSC markers across tumor types remains an active research challenge.

Not directly symptomatic; inferred from tumor behavior
Suspected when tumors recur after apparent complete response
Implicated in dormancy and late recurrences
Associated with chemotherapy and radiation resistance
Linked to metastatic potential

Tumor types with high recurrence after first-line therapy
