What is Tafamidis: Transthyretin Stabilizer Therapy?

Tafamidis is an oral drug that stabilizes transthyretin tetramers and prevents their dissociation into monomers and the formation of amyloid fibrils. The ATTR-ACT trial showed reductions in mortality and cardiovascular hospitalization.

It is indicated in both variant (hATTR) and wild-type (ATTRwt) cardiomyopathy. It is effective in NYHA class I–III patients and less effective in advanced disease.

It is given as 61 mg/day (tafamidis meglumine) or 80 mg/day (tafamidis free acid). The safety profile is generally favorable, with mild gastrointestinal symptoms as the most common side effect.
